      目的 探讨IL-1β对人真皮成纤维细胞(human dermal fibroblast,HDF)的细胞抑制率、促血管生成因子和炎症因子分泌的影响.方法 CCK8法检测不同浓度IL-1β对HDF的细胞抑制率,ELISA法检测3,10ng/mLIL-1β处理HDF 24h后上清中血管内皮生长因子(VEGF)、成纤维细胞生长因子(FGF)、IL-6的表达.结果 0.3、1、3、10ng/mLIL-1β对HDF的抑制率分别为(1.22±0.78)%,(3.72±1.21)%,(9.10±2.60)%,(14.42±3.36)%.3、10ng/mLIL-1β作用于HDF 24h后,两组上清的VEGF、FGF水平较空白对照组均升高(FGF P<0.05,VEGF P<0.01),3,10ng/rnl IL-1β两个浓度组间无统计学差异.3、10ng/mLIL-1β两组HDF上清中IL-6浓度较空白对照组均增加(P<0.05,P<0.001),且10ng/mLIL-1β组较3ng/mL组IL-6浓度更高,有统计学差异(P<0.001).结论 IL-1β可促进HDF分泌VEGF、FGF和IL-6,提示IL-1β可能通过促进成纤维细胞分泌促血管生成和炎症因子,进而促进肿瘤生成、浸润和转移.

      Objective To explore the role of interleukin-1β (IL-1β) in inhibiting the growth of human dermal fibroblast (HDF) and in promoting HDF secretion of angiogenic and inflammatory factors.Methods CCK8 was applied in detecting the growth inhibiting rate of HDF treated with IL-1β of different concentrations while ELISA was applied in detecting the expression levels of vascular endothelial growth factor (VEGF),fibroblast growth factor (FGF) and IL-6 in HDF treated with 3 and 10ng/mL IL-1β for 24 hours.Results The growth inhibiting rate of HDF treated with 0.3ng/mL IL-1β,1 ng/mL IL-1β,3 ng/mL IL-1β and 10ng/mL IL-1β was (1.22±0.78) %,(3.72±1.21) %,(9.10±2.60) % and (14.42±3.36) % respectively;the expressions of VEGF and FGF in 3 and 10ng/ml IL-1[β group increased significantly and were higher than those in blank control group (VEGF P<0.01,FGF P<0.05) while there existed no statistical difference in the expressions between 3 and 10ng/ml IL-1β group;the expressions of IL-6 in 3 and 10ng/ml IL-1β group were higher than that in blank control group (P<0.05,P<0.001) while the expression of IL-6 in 10ng/ml IL-1β group was much higher than that in 3ng/ ml IL-1β group (P<0.001).Conclusions IL-1β can promote HDF secretion of VEGF,FGF and inflammatory cytokine IL-6,which indicates that IL-1β may promote the tumorigenesis,invasion and metastasis..
