益气化聚方对早期糖尿病肾病大鼠尿微量白蛋白及细胞因子的影响

      目的 探讨益气化聚方对2型糖尿病早期肾病大鼠尿微量白蛋白及肾病蛋白(nephrin)、血管内皮生长因子受体2(VEGFR2)蛋白表达的影响.方法 建立2型糖尿病大鼠早期肾病模型,随机分为对照组、模型组、益气化聚方组、氯沙坦组.给药8周后检测各组空腹血糖、尿微量白蛋白/尿肌酐、24 h尿液微量白蛋白排泄率,观察肾组织病理学改变,检测nephrin、VEGFR2的表达及VEGFR2酪氨酸磷酸化(p-VEGFR2 (Tyr951)、p-VEGFR2 (Tyr1175》)水平.结果 与模型组比较,益气化聚方明显降低2型糖尿病早期肾病大鼠尿微量白蛋白/尿肌酐(P＜0.01)、24 h尿微量白蛋白排泄率(P＜0.05)及空腹血糖水平(P＜0.01),减轻肾小球基底膜增厚(P＜0.05)及细胞外基质增生(P＜0.01),上调肾组织nephrin表达(P＜0.01),下调VEGFR2蛋白表达及其酪氨酸磷酸化p-VEGFR2 (Tyr951)、p-VEGFR2 (Tyr1175)水平(P＜0.01).结论 益气化聚方可减轻2型糖尿病早期肾病大鼠尿微量白蛋白,其机制可能与调控nephrin、VEGFR2蛋白表达及磷酸化有关.

      Objective To investigate the effect of Yiqi Huaju Formula on microalbuminuria and the expressions of nephrin and vascular endothelial growth factor receptor 2 (VEGFR2) in rats with early type 2 diabetic nephropathy.Methods Early diabetic nephropathy models were built up in rats with type 2 diabetes and then the rats were randomly divided into 4groups:control group,model group,Yiqi Huaju Formula group and Losartan group;after the administration of Yiqi Huaju Formula or Losartan for 8 weeks,fasting serum glucose,urinary microalbumin creatinine ratio (ACR) and 24h urinary albumin excretion rate (UAER) of the rats in each group were detected,and the histopathologic changes were observed;the expressions of nephrin,VEGFR2,Phospho-VEGFR2 (Tyr951) and Phospho-VEGFR2 (Tyr1175) in kidney tissues of the rats in the 4 groups were studied.Results The urinary microalbumin creatinine ratio (ACR),24h urinary microalbumin excretion rate (UAER) and the level of fasting serum glucose of the rats in Yiqi Huaju Formula group decreased much more than those of the rats in other groups (P＜0.01,P＜0.05 and P＜0.01),the glomerular basement membrane thickening and the exracellular matrix proliferation were more alleviated (P＜0.01,P＜0.05),and the expression of nephrin in kidney tissues was up-regulated while the expressions of VEGFR2,Phospho-VEGFR2 (Tyr951) and Phospho-VEGFR2 (Tyr1175) were down-regulated (P＜ 0.01).Conclusions Yiqi Huaju Formula can alleviate urinary microalbumin of the rats with early type 2 diabetic nephropathy,the mechanism may be related to the regulation of expressions of nephrin,VEGFR 2,p-VEGFR2 (Tyr951) and p-VEGFR2 (Tyr1175).
