| 目的 体外评价血管生成素样蛋白-4(ANGPTL4)对血管通透性的影响及可能的机制.方法 在正常氧分压和低氧分压条件下,向体外培养的人脐静脉内皮细胞(HUVECs)中加入不同浓度的ANGPTL4(空白对照,0.625μg/mL、1.25 μg/mL、2.5 μg/mL和5μtg/mL),用二室弥散法测定内皮细胞的通透性;用免疫荧光法检测β-Catenin,ZO-I的分布.结果 同对照组相比,0.625 μg/mL、1.25 μg/mL、2.5 μg/mL和5μg/mL ANGPTL4组通透性均显著增加(P<0.05);低氧状态同相对应的正常氧分压状态相比,通透性增加更明显.ANGPTL4促进β-Catenin从细胞膜向细胞核转移,增加了细胞连接的间隙;缺氧状态更明显.结论 ANGPTL4能增加人脐静脉内皮细胞通透性,效应呈浓度依赖;缺氧状态下效果更强.ANGPTL4的作用与内皮细胞间连接破坏和致密连接受损可能相关. |

| Objective To evaluate the effect of angiopoietin-like 4 (ANGPTL 4) on vascular permeability and the mechanisms.Methods Under normal oxygen or hypo-oxygen condition,human umbilical vein endothelial cells (HUVEC)were cultured in medium containing ANGPTL 4 at different concentrations and the permeability was detected as diffusion of FITC-BSA across HUVEC monolayer and distribution of [β-Catenin and ZO-1 by immunoflurescence assay.Results At any working concentration from 0.625 μtg/mL to 5 μg/mL,ANGPTL4 increased the HUVEC monolayer permeability,especially under hypo-oxygen condition (P<0.05).ANGPTL4 promoted translocation of β-Catenin from cell surface to nucleus,and enlarged intercellular gap,which was accentuated under hypo-oxygen condition.Conclusions ANGPTL4 can increase HUVEC permeability effectively depending on concentration degree,especially under hypo-oxygen condition.This effect may be correlated to the interruption of intercellular connection. |
