      目的 探讨白细胞介素-18受体(Interleukin-18 Receptor,IL-18R)在脑出血中的作用,评估其作为脑出血潜在生物标志物的可能性,并揭示其在神经炎症和神经损伤中的潜在机制.方法 选用野生型C57BL/6小鼠通过自体血注射进行脑出血造模,应用CT成像、神经功能评分、拉力测试、足错误测试对造模结果进行评估.采用Western blot技术检测造模前后脑组织中的IL-18R表达情况,免疫荧光共定位追踪IL-18R造模前后的定位.结果 CT成像显示造模后的血肿存在;与假手术组相比,造模组神经功能评分降低,拉力测试的拉力数值降低、足错误的侧差增大,脑出血造模后的IL-18R表达增加,差异均有统计学意义(P<0.05);造模前后的IL-18R都与神经元产生共定位.结论 IL-18R可能在小鼠脑出血后的继发神经功能损伤中发挥重要作用;脑出血模型中IL-18R的表达显著增加,并与神经元共定位,提示其可能参与神经炎症机制;IL-18R有潜力作为脑出血的生物标志物,进一步研究其机制将可能为脑出血的治疗提供新策略.

      Objective To explore the role of interleukin-18 receptor(IL-18R)in intracerebral hemorrhage(ICH),evaluate its potential as a potential biomarker for ICH,and reveal its underlying mechanism in the neuroinflammation and nerve injury.Methods Wild type C57BL/6 mice were used to establish an ICH model through autologous blood injection.CT ima-ging,nerve function score,grip strength test and foot-fault test were used to evaluate the modeling results.Western blotting was used to detect the expression of IL-18R in brain tissue before and after the modeling.The immunofluorescence co-localiza-tion was used to track the localization of 1L-18R before and after the modeling.Results CT imaging showed the presence of hematomas after modelling.Compared with the-sham surgery group,the nerve function score of the modeling group decreased,tension value in grip strength test decreased,bilateral difference in foot-fault test increased,and expression of IL-18R in-creased.IL-18R co-localized with neurons both before and after modelling.Conclusion IL-18R may play an important role in the secondary neurological impairment after ICH in mice.The expression of IL-18R significantly increases in the ICH model and IL-18R exhibits co-localization neurons,suggesting that IL-18R may be involved in the mechanism of neuroinflammation and has the potential to serve as a biomarker for ICH.Further research on its mechanism may provide new strategies for the treatment of ICH.
