      目的 观察降脂胶囊对ApoE-/-小鼠动脉粥样硬化模型血脂、主动脉粥样硬化及TLR4/MyD88/NF-κB信号通路中关键蛋白及炎症因子的影响,探讨降脂胶囊治疗动脉粥样硬化的作用机制.方法 将ApoE-/-小鼠随机分为标准饮食(CON)和高脂饮食组(HFD).HFD组持续12周制备动脉粥样硬化模型.然后将高脂饮食喂养小鼠进一步分为模型组,阿托伐他汀(4.11 mg·kg-1)组,低剂量降脂胶囊(0.62 g·kg-1)组,高剂量降脂胶囊(1.24 g·kg-1)组,给药12周.取主动脉进行HE染色,检测血脂TG、TC、HDL-c和LDL-c水平.采用Elisa及Western blot方法检测炎症因子TNF-α、IL-1β、ICAM-1和MCP-1水平,采用Western blot方法检测TLR4、MyD88、NF-κBp65蛋白表达水平.结果 降脂胶囊降低ApoE-/-小鼠AS模型血脂水平,降低炎症因子TNF-α、IL-1β、ICAM-1、MCP-1水平及TLR4、MyD88、NF-κBp65蛋白表达.结论 降脂胶囊通过抑制TLR4/MyD88/NF-κB通路中关键蛋白及炎症因子的表达水平抑制AS的炎症反应.

      Objective To observe the effects of Jiangzhi capsule on blood lipid,aortic atherosclerosis(AS),key pro-teins in TLR4/MyD88/NF-κB signaling pathway and inflammatory factors in an ApoE-/-mouse AS model,and explore its mechanism in the treatment of atherosclerosis.Methods ApoE-/-mice were randomly divided into standard diet group(con-trol group)and high-fat diet group(HFD group).The HFD group was fed with high-fat diet for 12 weeks to establish an AS model.Subsequently,the mice in the HFD group were further subdivided into model group,atorvastatin group(4.11 mg/kg),low-dose Jiangzhi capsule group(0.62 g/kg),and high-dose Jiangzhi capsule group(1.24 g/kg).The duration of ad-ministration was 12 weeks.Aortic tissues were collected for hematoxylin-eosin(HE)staining,and blood lipid levels(TG,TC,HDL-c,and LDL-c)were measured.Enzyme-linked immunosorbent assay(ELISA)and Western blot were used to meas-ure the levels of inflammatory factors(TNF-α,IL-1 β,ICAM-1,MCP-1),and Western blot was employed to measure protein expression levels of TLR4,MyD88 and NF-κBp65.Results Jiangzhi capsule significantly decreased the levels of blood lipid level,inflammatory factors(TNF-α,IL-1 β,ICAM-1,MCP-1)and expression of TLR4,MyD88,and NF-κBp65 proteins in the AS model of ApoE-/-mice.Conclusion Jiangzhi capsule can inhibit the inflammatory response of AS by inhibiting the expression levels of key proteins in TLR4/MyD88/NF-κB pathway and inflammatory factors.
