目的 基于网络药理学结合动物实验,阐明黄芪在干预哮喘过程中的分子机制,并评估其在免疫衰老背景下的潜在治疗应用价值.方法 通过网络药理学分析,筛选黄芪的活性成分及其潜在靶点,结合基因数据库进行靶点预测与交集分析,构建成分-靶点-通路网络,进一步利用蛋白质互作网络(PPI)进行分析.随后,通过动物实验建立屋尘螨(HDM)诱导的哮喘小鼠模型,评估黄芪对肺功能、气道炎症及免疫反应的干预效果.结果 网络药理学分析揭示,黄芪的活性成分主要通过调节免疫细胞功能、炎症反应及气道重塑等多个信号通路发挥作用,关键靶点包括IL-4、IL-5、IL-13、IL-17A等.动物实验显示,黄芪干预显著改善哮喘小鼠的肺功能,减少气道炎症细胞浸润,抑制IL-4、IL-5、IL-13等Th2型细胞因子表达,下调IL-17A、IL-17F等Th17型细胞因子水平,且未观察到显著的肝肾毒性.结论 黄芪可能通过多靶点、多通路调节免疫炎症反应,显著改善老年哮喘模型小鼠的肺功能和气道炎症,具有成为老年哮喘有效治疗策略的潜力.该研究为黄芪在老年哮喘治疗中的应用提供理论依据,并为中药在老年免疫性疾病治疗中的应用提供新思路. |

Objective To elucidate the molecular mechanism of Astragalus(Huangqi)in the intervention of asthma based on network pharmacology combined with animal experiments,and evaluate its potential therapeutic application value in the immunosenescence.Methods Through network pharmacology analysis,active component and potential targets of Astraga-lus were screened.Target prediction and intersection analysis were carried out in combination with the Gene Database to con-struct a component-target-pathway network,and protein-protein interaction(PPI)network was further used for analysis.Subse-quently,a mouse model of asthma induced by house dust mite(HDM)was established for animal experiments.The interven-tion effects of Astragalus on lung function,airway inflammation and immune response were evaluated.Results Network phar-macology analysis revealed that the active components of Astragalus mainly exert their effects through regulating multiple signa-ling pathways such as immune cell function,inflammatory response,and airway remodeling.Key targets included IL-4,IL-5,IL-13,IL-17A,etc.Animal experiments showed that Astragalus intervention significantly improved the lung function of asthma mice,reduced the infiltration of airway inflammatory cells,inhibited the expression of Th2-type cytokines such as IL-4,IL-5,and IL-13,and down-regulated the levels of Th17-type cytokines such as IL-17A and IL-17F.No significant hepatotoxicity or nephrotoxicity was observed.Conclusion Astragalus may regulate immune inflammatory responses through multiple targets and pathways,significantly improve lung function and airway inflammation in elderly asthma model mice.It has the potential to become an effective therapeutic strategy for elderly asthma patients.This study provides a theoretical basis for the application of Astragalus in the treatment of asthma in the elderly and offers new ideas for the application of traditional Chinese medicine in the treatment of immune diseases in the elderly. |
